A simple, fast, and orientation-controllable technology for preparing antibody-modified liposomes

Modification with antibodies is a useful strategy for the delivery of nanoparticles to target cells. However, complexity required chemical modifications makes them time-consuming and low efficiency, orientation antibody challenging control. To develop simple, fast, effective, orientation-controllable technology, we employed staphylococcal protein A, which can bind Fc region antibodies, as tool conjugating nanoparticles. Specifically, modified C-domain dimer A contain lysine cluster create molecule, DPACK, that would electrostatically anionic liposomes. Using this protein, antibody-modified liposomes be prepared in 35 min two steps: (1) interaction DPACK (2) an DPACK-modified Binding efficiencies IgG were 75% 72–84%, respectively. Uptake anti-epidermal growth factor receptor (EGFR) via by EGFR-expressing cancer cells was significantly higher than unmodified liposomes, accumulated tumors colocalized EGFR. This effective technology preparing will active targeting drug delivery.
